1. 洗滌緩沖液的制備與儲(chǔ)存
手冊(cè)中明確要求HDL重懸緩沖液需新鮮制備且不可儲(chǔ)存。對(duì)于1×HDL洗滌液，我個(gè)人建議每次新鮮配制，但將其置于4°C儲(chǔ)存直至使用是可行的，無需過度擔(dān)憂。
2. PBS配方建議
在此步驟中，您可根據(jù)下游應(yīng)用需求選擇適合的緩沖液進(jìn)行透析。目前未提及具體PBS配方（如pH、離子強(qiáng)度、是否含Ca2?/Mg2?或防腐劑如疊氮鈉）的強(qiáng)制要求，但需確保其與后續(xù)實(shí)驗(yàn)兼容。
3. 透析膜選擇
對(duì)于貨號(hào)Cat# STA-607，透析方案可采用任意透析方法。例如，可使用賽默飛世爾的Slide-A-Lyzer MINI透析裝置。我們通常推薦使用10 kDa截留分子量（MWCO）的透析膜，但其他尺寸也可接受。操作流程需遵循說明書，一般步驟如下：
• 用300倍樣本體積的緩沖液透析2-4小時(shí)；
• 更換緩沖液，繼續(xù)透析2-4小時(shí)；
• 再次更換緩沖液，過夜透析；
• 建議在4°C下進(jìn)行透析以保持樣本穩(wěn)定性。
4. 詳細(xì)透析流程
請(qǐng)參考上述第3題的答案（透析時(shí)長(zhǎng)、緩沖液更換頻率及溫度要求均已涵蓋）。
5. 透析后樣本情況
• 最終體積：Cat# STA-607的典型產(chǎn)量約為7 ml（來自人血清或血漿樣本）；
• 蛋白濃度范圍：純化后HDL的濃度約為4500 µg/ml。
6. 透析后樣本處理
需注意，Cat# STA-607提供的溶液并非無菌，因此我們從未對(duì)純化后的HDL進(jìn)行過過濾除菌。理想情況下，純化后的脂蛋白可長(zhǎng)期儲(chǔ)存于4°C；若樣本含50%甘油，則可儲(chǔ)存于-20°C。關(guān)于分裝體積，我們無法給出具體建議，需客戶根據(jù)下游應(yīng)用自行決定。
7. 蛋白濃度測(cè)定方法推薦
手冊(cè)未明確蛋白濃度測(cè)定方法，建議客戶根據(jù)下游應(yīng)用選擇合適方法。我們通常采用BCA assay測(cè)定總蛋白濃度。
8. 總蛋白濃度與HDL濃度的等效性
遺憾的是，我們從未將BCA assay測(cè)定的濃度與ELISA結(jié)果進(jìn)行過比較（僅通過ELISA確認(rèn)ApoB的存在）。因此，無法提供校正方法或參考標(biāo)準(zhǔn)。關(guān)于殘留的非HDL蛋白，我們通過SDS-PAGE評(píng)估純度。
9. ApoM穩(wěn)定性
很遺憾，我們從未使用Cat# STA-607評(píng)估過ApoM的穩(wěn)定性（包括其在硫酸葡聚糖沉淀、離心、重懸、洗滌及透析過程中的結(jié)合情況），也無相關(guān)數(shù)據(jù)證明其未解離。
10. S1P穩(wěn)定性
同樣，我們從未評(píng)估過ApoM或S1P在純化過程中的穩(wěn)定性（如離心、渦旋/重懸、透析等步驟是否會(huì)導(dǎo)致S1P與ApoM解離或部分丟失）。
11. HDL顆粒完整性
該試劑盒旨在維持HDL顆粒的天然結(jié)構(gòu)（包括脂質(zhì)雙層及載脂蛋白組成），但我們從未對(duì)其結(jié)構(gòu)或功能進(jìn)行過驗(yàn)證（如電子顯微鏡、天然凝膠電泳等數(shù)據(jù)）。
12. ApoM-S1P-HDL的沉淀行為
我們從未評(píng)估過ApoM或S1P與HDL的結(jié)合情況，因此無法確定ApoM-S1P-HDL是否會(huì)與總HDL共同沉淀，或是否存在選擇性丟失的風(fēng)險(xiǎn)。
13. 最終產(chǎn)物的組成
按照手冊(cè)操作獲得的最終產(chǎn)物是否為“總HDL"？是否包含攜帶ApoM的HDL亞群（即ApoM陽性HDL）？若有，是否有參考數(shù)據(jù)（如文獻(xiàn)或內(nèi)部驗(yàn)證）說明其在總HDL中的比例？
很遺憾，我們從未評(píng)估過ApoM或S1P的相關(guān)情況，無法回答此問題。此外，關(guān)于純化產(chǎn)物中是否殘留其他脂蛋白（如LDL、VLDL）或血漿蛋白，我們通過SDS-PAGE評(píng)估純度。
14. 直接兼容性
純化后的HDL產(chǎn)物（透析后）是否可直接用于ApoM免疫沉淀？是否需要額外的緩沖液交換或純化步驟（如脫鹽、超濾）？
很遺憾，我們從未評(píng)估過ApoM或S1P的相關(guān)情況，無法給出建議。
15. 殘留干擾成分
純化后的HDL產(chǎn)物中是否有殘留成分（如硫酸葡聚糖、Tris、沉淀試劑等）可能干擾抗體-抗原結(jié)合？這些殘留是否會(huì)影響免疫沉淀效率？
通過使用硫酸葡聚糖去除液并進(jìn)行透析，可去除大部分干擾成分。
16. 抗體推薦
是否有經(jīng)過驗(yàn)證的ApoM免疫沉淀抗體推薦（如克隆號(hào)或目錄號(hào)）？關(guān)于抗體濃度、孵育條件或洗脫方法，是否有建議？
很遺憾，我們從未評(píng)估過ApoM或S1P的相關(guān)情況，無法提供推薦。
17. 產(chǎn)物組成細(xì)節(jié)
純化后的HDL產(chǎn)物中已知成分有哪些（如載脂蛋白種類、脂質(zhì)組成、緩沖液成分）？是否有批間一致性數(shù)據(jù)？
我們通過SDS-PAGE評(píng)估純度，但未開展更深入的成分分析或批間一致性研究。
18. 直接純化產(chǎn)品及技術(shù)服務(wù)
